The nuclear lamina consists of two components lamins and nuclear lamin associated membrane proteins. Loss of specific lamins does not affect protein levels of other lamins we analyzed the function of specific lamins in nuclear shape stability and stiffness in wild type mefs and in mefs lacking lamin a designated lamin c only or lmna lco lco cells in mefs deficient in both lamins a and c lmna and in mefs expressing a severely truncated mutant form of lamin b1 lmnb1. Left collagen and lamin a levels scale with e consistent with matching tissue stress to nuclear mechanics right matrix stiffness in tissue culture increases cell tension and stabilizes lamin a regulating its own transcription and that of stress fiber genes enhancing.
